Excited for the preprint of my first, first-author paper! This was such a talented group of scientists to work with. Especially my co-first author and fellow graduate student, Juan Carlos Martinez Cervantes, and my PI, Dr. Haiyan He. Expansion microscopy (ExM) is a novel tool developed in 2015 designed to overcome the diffraction limit on easily accessible microscopes such as the confocal, with up to 70nm resolution. Despite the advantages ExM offers, it is a cumbersome process with prolonged wait times. Here, we used a specialized microwave to speed up the ExM process. Surprisingly, not only did this maintain the superior resolution of the original ExM protocol, but it also increased the expansion factor. We first did this in Xenopus laevis tadpoles and then collaborated with the Cervantes lab as they reproduced these results in Drosophila melanogaster fruit flies. If you are interested in asking scientific questions about subcellular resolution while using conventional microscopes, this protocol may be for you!

On this day in science Erwin Neher was born. He was part recipient of the 1991 Nobel Prize for his work on single-ion channels in biological cells Erwin Neher and Bert Sakmann’s patch-clamp technique is based on an exquisitely simple idea. An extremely fine glass pipette with a very small opening is used to make contact with a tiny area, or patch, of the cell’s outer membrane that, with some luck, will contain only a single ion channel. Applying a small amount of suction through the pipette forms a seal tight enough such that ions can only flow from the channel into the pipette. By fitting the pipette with a highly sensitive electrode, Neher and Sakmann could record every minute change in current produced as ions flow through the clamped channel into the pipette. Throw back to last week at LINXS Institute of advanced Neutron and X-ray Science where Claire L Lyons heard talks from Swedish and international researchers and infrastructures about how to use neutrons for drug discovery, design, and formulation research. Neutrons can be used for structure analysis, contrast imaging and multi-compositional analysis. This is useful for looking at the role of hydrogen bonding and drug interactions, liquid-aqueous interfaces, and lipid nanocarriers for mRNA delivery as well as interactions at lipid bilayers, all important for various stages of drug design and development. We heard how these experiments will be possible at European Spallation Source ERIC in the future using SANS (small angle neutron scattering), reflectometry and macromolecular crystallography. An exciting announcement was that workshops demonstrating early science at ESS will start this year, for example at Swedish Neutron Week which is on this month in Lund. https://neutronweek.site/ Already available at ESS is the Deuteration and Macromolecular Crystallisation (DEMAX) which offers both chemical and biological deuteration, which are necessary for working with many neutron experiments. 𝗗𝗶𝗱 𝗬𝗼𝘂 𝗞𝗻𝗼𝘄 𝗥𝗼𝘀𝗮𝗹𝗶𝗻𝗱 𝗙𝗿𝗮𝗻𝗸𝗹𝗶𝗻 𝗣𝗿𝗼𝗱𝘂𝗰𝗲𝗱 𝘁𝗵𝗲 𝗙𝗶𝗿𝘀𝘁 𝗖𝗹𝗲𝗮𝗿 𝗫-𝗿𝗮𝘆 𝗜𝗺𝗮𝗴𝗲𝘀 𝗼𝗳 𝗖𝗿𝘆𝘀𝘁𝗮𝗹𝗹𝗶𝘇𝗲𝗱 𝗗𝗡𝗔? Today, Rosalind Franklin was born 103 years ago today. Franklin is best known for her work on the #XRay diffraction images of #DNA with the sugars and phosphates on the outside and the bases on the inside. Her work led to the discovery of the DNA double helix structure for which Francis Crick, James Watson and Maurice Wilkins shared the “Nobel Prize in Physiology or Medicine” in 1962. Although she is often dubbed the "forgotten heroine" and "feminist icon" of #molecularbiology, the fact that Franklin never received the Nobel Prize, however, was less the result of concealing her scientific achievements as a woman than of her early death: After being diagnosed with cancer in the autumn of 1956, she only had a good year and a half left until she died at the age of 37. Franklin also led pioneering work on the molecular structures of #viruses. After her death her colleague Aaron Klug continued her research, winning the Nobel Prize in Chemistry in 1982 for his development of crystallographic electron microscopy and his structural elucidation of biologically important nucleic acid-protein complexes. Some of the most state-of-the art scientific analytical methods that belong to ProtaGene's service portfolio to support our clients' biotherapeutics projects are based on the research of the gifted scientist Franklin, whose achievements we commemorate today.
