Summary Virtually all cells use energy-driven, ion-specific membrane pumps to maintain large transmembrane gradients of Na+, K+, Cl−, Mg++, and Ca++, but the corresponding evolutionary benefit remains unclear. We propose that these gradients enable a dynamic and versatile biological system that acquires, analyzes, and responds to environmental information. We hypothesize that environmental signals are transmitted into the cell by ion fluxes along pre-existing gradients through gated ion-specific membrane channels. The consequent changes in cytoplasmic ion concentration can generate a local response or orchestrate global/regional cellular dynamics through wire-like ion fluxes along pre-existing and self-assembling cytoskeleton to engage the endoplasmic reticulum, MITOCHONDRIA, and nucleus. https://lnkd.in/gmyyS2-i

I present the results of our recent work on the interaction between bis(8-hydroxiquinolinate)oxidovanadium(IV) and RNase A. Unespectedly, the metal complex is transformed by interacting with residues close to the coordination site on the protein surface. This is the first time I see a protein acting like an enzyme towards a metal complex, producing a species that, otherwise, would not be present in solution. https://lnkd.in/eJtJia_C

I have seen thousands of crystal structures... 🤔 This one makes me wonder, how much of the complexity of molecular interactions waits to be uncovered. Look at this water molecule and its coordination! And how glutamate is interacting with the water molecule and lysine. High-resolution is so important to capture the local dynamics of proteins... #crystallography #drugdiscovery
