Dana Holzinger et al, PROGEN recently presented results of their blind study comparing various methods of AAV capsid titer on their newly released reference material. The Gator BLI assay showed very low variation resulting in CVs of around 5%. Each method showed a serotype-dependent average deviation compared to ELISA. Overall, Gator BLI deviation across serotypes compared to ELISA was only 16%. High specificity AAV genome titer is traditionally done using qPCR and digital PCR; while these methods deliver high quality data, they are slow, tedious and not capable of strand specific titer. GeneSwift’s strand specific titer capability enables genome integrity of self-complementary AAV (scAAV).
