Adam Crowe (Cytiva) recently gave a wonderful #webinar on the correlation between lipid purity and mRNA lipidation. In particular, mRNA adducts can result due to the breakdown products of N-oxide impurities. The ionizable lipids used in LNPs are especially susceptible to forming N-oxide impurities. Targeted degradation has emerged as a powerful therapeutic modality. Relying on “event-driven” pharmacology, proteolysis targeting chimeras (PROTACs) can degrade targets and are superior to conventional inhibitors against undruggable proteins. Scientists from University of Pennsylvania present an engineered bioPROTAC template able to complex with cationic and ionizable lipids via electrostatic interactions for cytosolic delivery. Michael J Mitchell et al. continue to innovate and impress with their new single step #LNP formulation technique ("click-like chemistry"). They have also found a way to lojack and track these particles to better target specific organs. "We've developed what we call an amidine-incorporated degradable (AID) lipid, a uniquely structured biodegradable molecule," Mitchell says. "Think of it as an easy-to-build custom mRNA vehicle with a body kit that informs its navigation system. By adjusting its shape and degradability, we can enhance mRNA delivery into cells in a safe manner. By adjusting the amount of the AID lipid that we incorporate into the LNP, we can also guide it to different organs in the body, much like programming different destinations into a GPS."
