It is well-established that cardiovascular disease is the leading cause of death worldwide. That is why continued research into this topic, and how survival rate following heart attack and heart failure can be increased, is important. In this study, Laura Vettori et al has evaluated the effect of silk fibroin (SF) on cardiac bioink formulations with cardiac spheroids, evaluating whether this could lead to improvements in the desired behavior from printed cardiac tissue. Building on previous work from the group, where alginate-gelatin (Alg-Gel) hydrogels were used to create beating patches of heart tissue, Vettori et al looked at how incorporating SF would impact the structural and elastic properties of alginate-gelatin hydrogels, as well as the printability and durability of the hydrogels. Overall, the study showed that adding 1% SF to Alg-Gel hydrogels improved the hydrogels durability over 28 days, and did not affect cell viability, but improved contractile activity. Meanwhile, increasing the concentration of SF to 2% resulted in strong 3D architectures and a reduction of pore size, which had a negative impact on cell viability. This study showcases the promise of SF for cardiac tissue bioprinting.
